Amaç: Giardia duodenalis, kuzularda verim kaybına yol açan ve asemblaj diye adlandırılan alt tipleri bulunan ciddi bir protozoer hastalıktır. Asemblaj A ve B hem insan hem de hayvanlarda bulunabilen zoonoz türlerdir. G. duodenalis özellikle genç bireylerde diyare ile seyreden gastrointestinal sorunlara ve hatta ölümlere sebep olabilmekte ve hayvan yetiştiriciliğinde önemli ekonomik kayıplara yol açmaktadır. Özellikle diyareli hayvanlarda varlığının araştırılması önemlidir. Bu sebeplerden yola çıkılarak planlanan bu çalışmada Kırıkkale bölgesindeki kuzularda bulunan G. duodenalis'in yaygınlığını belirlemek amaçlanmıştır. Gereç ve Yöntemler: Bu çalışmada Kırıkkale ilinde değişik işletmelerden 79 adet kuzudan dışkı toplandı. Alınan örneklerden doymuş çinko sülfat flotasyon (ZnSO4; %33) yöntemi kullanılarak mikroskobik olarak incelemeler yapıldı. Daha sonra 79 kuzu dışkısından DNA ekstraksiyonu yapılarak ve elde edilen DNA'ların nanodropla konsantrasyonu ölçülerek bu DNA'lardan G. duodenalis spesifik primerler kullanılarak PCR (Polymerase Chain Reaction) testi yapıldı, Daha sonra jel görüntüleme sisteminde görüntüleri elde edildi. Bulgular: Mikroskobik inceleme sonucunda 79 örneğin 13 (%16,45)'ünde G. duodenalis kistleri görüldü. 79 kuzu dışkısına PCR yapıldı ve 16 (%20,25)'sında 511 bp büyüklüğünde bantlar elde edildi. Sonuç: Sonuç olarak Kırıkkale bölgesinde ki kuzularda G. duodenalis'in önemli bir sorun teşkil ettiği, uygun tedavinin uygulanmasının, koruyucu tedbirlerin alınmasının ve halk sağlığını korumak adına yetiştiricinin bu konuda bilinçledirilmesinin zorunlu olduğu, ayrıca sonraki çalışmalar ile bu yöredeki kuzularda G. duodenalis'in alt tiplerinin araştırılarak zoonoz potensiyele sahip asemblajların yayılımının belirlenmesinin faydalı olacağı anlaşılmıştır.

Objective: Giardia duodenalis is a serious protozoan disease that causes loss of yield in lambs and has subtypes called assemblage. Assemblage A and B are zoonoses that can be found in both humans and animals. G. duodenalis can cause gastrointestinal problems and even deaths with diarrhea especially in young individuals and cause significant economic losses in animal husbandry. It is important to investigate its presence, especially in diarrheal animals. In this study, which was planned based on these reasons; it was aimed to determine the prevalence of G. duodenalis in lambs in Kırıkkale region. Material and Methods: In this study, feces were collected from 79 lambs from different enterprises in Kırıkkale province. Microscopic examinations were carried out from the samples taken using saturated zinc sulfate flotation (ZnSO4; 33%) method. Then, DNA extraction was performed from 79 lamb feces and the concentration of DNA obtained with nanodrop was measured and PCR test was performed using G. duodenalis specific primers from these DNA, Then, images were obtained in the gel imaging system. Results: As a result of microscopic examination, G. duodenalis cysts were seen in 13 of 79 samples (16.45%). PCR was performed in 79 lamb feces and 511 bp bands were obtained in 16 (20.25%). Conclusion: Consequently, G. duodenalis is an important problem in lambs in Kırıkkale region, it is obligatory to apply appropriate treatment, take protective measures and raise the awareness of the breeder in order to protect public health, and also in subsequent studies, G. duodenalis in lambs in this region. It was clearly understood that it would be beneficial to determine the spread of assemblage with zoonotic potential by investigating subtypes.
