Amaç: Bu çalışmanın amacı, Türkiye'nin Van ilindeki Van kedilerinde, Giardia duodenalis prevalansının mikroskobik ve moleküler olarak belirlenmesidir. Gereç ve Yöntemler: Bu çalışmada, 2020-2021 yılları arasında Van bölgesindeki özel veteriner kliniklerine tanı amaçlı gelen, 0-2 yaş arası ishal, huzursuzluk, kilo kaybı gibi klinik belirtileri olan toplam 80 Van kedisinin dışkı örneği toplanmıştır. Toplanan her numune için kedinin cinsiyeti ve yaşı kaydedildi. Daha sonra numuneler, soğuk zincirde laboratuvara getirilerek inceleninceye kadar +4°C'de saklandı. Mikroskobik inceleme de Nativ-lugol yöntemiyle mikroskop altında X40'lik objektifte Giardia spp. kistleri yönünden incelendi. Mikroskobik incelemesi yapılan dışkı örneklerinin tamamından DNA ekstraksiyonu yapıldı. Elde edilen DNA'lar bir sonraki aşamalara kadar -20°C'de muhafaza edildi. Daha sonra ilgili primerlerle Nested PCR yapıldı. Ardından %1,5'lik agaroz jel hazırlanıp jel görüntüleme cihazında görüntüleri elde edildi. Bulgular: Mikroskobik inceleme sonucunda, 80 örneğin 17'inde (%21,25) Giardia spp. kistleri tespit edilmiştir. Nested PCR ile 80 örneğin 20'sinde (%25) G. duodenalis'e özgü 511 bp büyüklüğünde bantlar elde edildi. Sonuç: Sonuç olarak Nested PCR yönteminin mikroskobik incelemelere göre daha güvenilir olduğu bulundu. Ayrıca Van kedilerinde Giardiazisin yaygın olduğu ve koruma kontrolleri için gerekli önlemlerin alınması gerektiği sonucuna varıldı. Yapılan araştırmalardan ve literatür taramalarından anlaşılacağı üzere, kedilerde Giardiozis ile ilgili diğer türlere göre az çalışma mevcuttur. Bu çalışma, literatüre katkı sağlaması ile birlikte başka çalışmalara alt yapı hazırlar niteliktedir.

Objective: The aim of this study is to determine the microscopic and molecular prevalence of Giardia duodenalis in Van cats in Van province of Türkiye. Material and Methods: In this study, stool samples of 80 Van cats with clinical symptoms such as diarrhea, restlessness, weight loss aged 0-2 years, who came to the private veterinary clinics in the Van region for diagnostic purposes between 2020-2021, were collected The sex and age of the cat were recorded for each sample collected. Then the samples were brought to the laboratory on a cold chain and stored at +4°C until they were examined. Microscopic examination was also performed using the Native-lugol method under a microscope with an X40 lens in terms of Giardia spp. cysts. DNA extraction was performed from all stool samples that were examined microscopically. The obtained DNA was stored at -20°C until the next stages. Then, Nested PCR was performed with the relevant primers. Then, 1.5% agarose gel was prepared and images were obtained on the gel imaging device. Results: As a result of microscopic examination, Giardia spp. cysts were detected. By Nested PCR, bands of 511 bp specific for G. duodenalis were obtained in 20 (25%) of 80 samples. Conclusion: As a result, the Nested PCR method was found to be more reliable than microscopic examinations. In addition, it was concluded that Giardiasis is common in Van cats and that necessary precautions should be taken for protection controls. As can be understood from the research and literature reviews, there are few studies on giardiosis in cats compared to other species. This study, along with contributing to the literature, prepares the infrastructure for other studies.
