ÖZET
Amaç: Farklı cins Gram-negatif bakterilerin aynı ortamda üretilmesi sonucu oluşturulan biyofilm miktarlarında meydana gelebilecek farklılıklar ile bunun siprofloksasine olan duyarlılığa etkisinin araştırılması amaçlandı. Gereç ve Yöntemler: Escherichia coli, Klebsiella pneumoniae ve Pseudomonas aeruginosa standart suşlarının siprofloksasine olan duyarlılıkları disk difüzyon ve mikrodilüsyon yöntemiyle belirlendi. Ayrıca bu bakterilerle oluşturulan ikili-üçlü karışık kültürlerin de siprofloksasine duyarlılığı mikrodilüsyon yöntemiyle araştırıldı. Her bir suşun ayrı ayrı ve ikili-üçlü karışımlarıyla hazırlanan kültürlerin biyofilm oluşturma düzeyi siprofloksasin uygulanmadan önce ve sonra kristal viyole yöntemiyle test edildi. Bulgular: E. coli, K. pneumoniae ve P. aeruginosa için siprofloksasin minimum inhibitör konsantrasyonları sırasıyla 0,008 mg/L; 0,016 mg/L ve 0,250 mg/L olarak tespit edildi. Siprofloksasin inhibisyon zon çapları her üç bakteri için de 30 mm'nin üzerindeydi. Karışımlar için siprofloksasin minimum inhibitör konsantrasyonları, E. coli+K. pneumoniae kültüründe 0,016 mg/L, P. aeruginosa'nın diğer iki bakteri ile ikili ve üçlü karışımlarında ise 0,250 mg/L olarak tespit edildi. Siprofloksasin uygulamasından önce ve minimum inhibitör konsantrasyonlarının altındaki değerlerde hem tek hem karışık bakteri kültürlerinde güçlü düzey biyofilm oluşumu saptandı. Minimum inhibitör konsantrasyon ve üzerindeki değerlerde ise biyofilm oluşmadı veya zayıf biyofilm oluşumu gözlendi. Sonuç: Doğal koşullardaki gibi farklı bakterilerin bir araya gelmesiyle oluşturulan kültürlerdeki biyofilm düzeylerinin ve siprofloksasine olan duyarlılığın değişiminin araştırıldığı bu çalışmada, biyofilm oluşturan mikroorganizmalarla mücadeleye katkı sağlayabilecek bazı ön verilere ulaşıldığından biyofilmle ilişkili enfeksiyonların tedavisine yönelik benzer ve ileri çalışmalar için yol gösterici olacağı düşünülmektedir.

ABSTRACT
Objective: It was aimed to investigate the differences that may occur in the amount of biofilm formed due to growing different types of Gram-negative bacteria in the same environment and the effect of this on the susceptibility to ciprofloxacin. Material and Methods: The susceptibility of Escherichia coli, Klebsiella pneumoniae and Pseudomonas aeruginosa standard strains to ciprofloxacin were determined by disk diffusion and microdilution method. In addition, the susceptibilities of double-triple mixed cultures formed with these bacteria to ciprofloxacin were investigated by microdilution method. Biofilm formation levels of cultures prepared with separate and double-triple mixtures of each strain were tested by crystal violet method before and after ciprofloxacin application. Results: The minimum inhibitory concentrations of ciprofloxacin to E. coli, K. pneumoniae and P. aeruginosa were determined as 0.008 mg/L, 0.016 mg/L and 0.250 mg/L, respectively. The ciprofloxacin inhibition zone diameters were over 30 mm for all three bacteria. Minimum inhibitory concentrations of ciprofloxacin for the mixtures, E. coli+K. pneumoniae culture, it was determined as 0.016 mg/L, and 0.250 mg/L in double and triple mixtures of P. aeruginosa with the other two bacteria. Strong biofilm formation was detected in both single and mixed bacterial cultures before ciprofloxacin administration and at values below the minimum inhibitory concentrations. At the minimum inhibitory concentration and above, no biofilm was formed or weak biofilm formation was observed. Conclusion: In this study, which investigated the change in biofilm levels and susceptibility to ciprofloxacin in cultures formed by the combination of different bacteria as in natural conditions, it is thought that it will be a guide for similar and advanced studies on the treatment of biofilm-related infections, since some preliminary data that can contribute to the fight against biofilm-forming microorganisms have been reached.

4.2. People benefiting from the services provided by "Turkiye Klinikleri" and using the website can use the "SITE" only according to the law and only for personal reasons. Turkish Law will be applied in practicing, interpreting the hereby "Terms of Use" and managing the emerging legal relationships within this "Terms of Use" in case of finding element of foreignness, except for the rules of Turkish conflict of laws. Ankara Courts and Enforcement Offices are entitled in any controversy happened or may happen due to hereby contract.

9. CLOSING AND AGREEMENT

Hereby "Terms of Use" come into force when announced in the "SITE" by "Turkiye Klinikleri". The users are regarded to agree to hereby contract terms by using the "SITE".
